

Low biomass samples present a unique challenge in molecular biology workflows because they contain very limited amounts of target nucleic acids to begin with. Unlike high-input samples, there is often little to no opportunity to repeat an extraction if the initial isolation underperforms. In many cases, these samples are precious, hard to collect, or available only in extremely small volumes. Obtaining high-quality DNA for downstream applications such as metagenomic sequencing, microbial profiling, or whole genome sequencing can be difficult when starting material is inconsistent or scarce.
SeqCenter recently worked with a set of mouse urine samples with input volumes ranging from as little as 20 µL up to 500 µL. Urine is inherently a low-biomass sample type, often containing very low microbial abundance alongside compounds that can interfere with molecular workflows. The variability in sample volume added another layer of complexity, requiring cautious handling and protocol optimization to maximize DNA recovery. Because many of these samples were single-use collections, the extraction workflow had to be approached conservatively and methodically to avoid unnecessary sample loss. Through meticulous processing and extraction refinement, SeqCenter was able to achieve measurable DNA yields across the sample set, allowing successful progression into library preparation and sequencing.
